| 为构建红锥 Castanopsis hystrix 优树茎段组织培养体系, 突破红锥优良无性系工厂化育苗瓶颈,
研究在秋季干燥晴朗天气以红锥 22-3 号优树半木质化穗条为外植体, 采用单因素分析, 探讨不同基本培
养基和 6-BA 浓度对增殖效果的影响, 以及不同基本培养基、 不同 IBA 和蔗糖浓度对生根效果的影响。
结果表明, 红锥优树茎段外植体消毒方法为 75%酒精处理 5 s, 无菌水冲洗 1 次, 0. 1%升汞浸泡 5 min,
无菌水冲洗 3 次; 初代诱导培养基为 WPM+2. 0 mg·L

| To establish a tissue culture system for the stem segments of Castanopsis hystrix, dominant trees
and break through the bottleneck of factory seedling cultivation of excellent C. hystrix clones, this study used the
semi-lignified cuttings of the C. hystrix dominant tree 22-3 collected under dry and sunny weather in autumn as
explants. A single-factor experiment design was employed to investigate the effects of different basic media and
6-BA concentrations on the proliferation efficiency, as well as the effects of different basic media, IBA and sucrose concentrations on the rooting efficiency. The results showed that the optimal disinfection method for the
stem segment explants of C. hystrix dominant trees was treatment with 75% alcohol for 5 s, rinsed once with
sterile water, followed by immersion in 0. 1% mercuric chloride for 5 min, and than rinsed three times with
sterile water. The primary induction medium was WPM+2. 0 mg·L
