马尾松和湿地松内生真菌群落多样性差异分析

摘要:
为建立基于松树内生菌的松材线虫生防菌株资源库, 研究选取广东省韶关市曲江区 (24. 722 287°N, 113. 626 33°E) 松材线虫病重度感病松林中的 15 a 生马尾松 Pinus massoniana 和湿地松 P. elliottii, 每个树种采集 3 根中部感病枝条, 通过 Illumina NovaSeq-PE250 平台对真菌 ITS1 区进行双端测序, 经 QI-IME2 流程质控后, 使用 UNITE 数据库进行物种注释, PICRUSt2 预测功能基因。 采用 α-多样性 (Shan-non / Chao1 指数) 和 β-多样性 (PCoA 分析) 评估群落差异, LEfSe 算法鉴定标志性菌群, 构建抗病相关 真菌资源库。 结果显示, 马尾松内生真菌的物种丰富度高于湿地松, 但 α 和 β 分析显示两者内生真菌多 样性差异不显著。 两个树种的内生真菌主要由子囊菌门 Ascomycota 和担子菌门 Basidiomycota 的真菌组 成, 湿地松中子囊菌门的相对丰度高于马尾松。 在属的水平上, 马尾松与湿地松的内生真菌种类差异明 显, 马尾松的子囊菌门座囊菌纲中的 Neoheleiosa 属和半知菌门丝孢纲的 Catanulostroma 属数量明显少于 湿地松, 而子囊菌门腔孢纲的 Paraconiothyrium 属数量明显多于湿地松。 马尾松和湿地松内生真菌参与的 代谢通路无明显差异, 但在 Metabolic Clusters 的二级代谢通路 phospholipases 中, 与湿地松相比, 马尾松 内生真菌的富集水平显著上调 (P<0. 001)。

Difference Analysis of Endophytic Fungi Community Diversity in Pinus massoniana and P. elliottii

Abstract:
To construct a microbial strain resource bank based on pine endophytes for controlling pinewood nematodes (Bursaphelenchus xylophilus), 15 -year -old Pinus massoniana and P. elliottii trees were selected from a severely infected pine forest in Qujiang District, Shaoguan City, Guangdong Province (24. 722 287°N, 113. 626 33°E). For each tree species, three diseased middle branches were collected. Fungal ITS1 regions were subjected to paired-end sequencing using the Illumina NovaSeq-PE250 platform. After quality control via QIIME2 pipeline, species annotation was performed with the UNITE database, and functional gene prediction utilized PICRUSt2. Community differences were assessed through α diversity (Shannon / Chao1 indices) and β diversity (PCoA analysis), while biomarker taxa were identified via the LEfSe algorithm to establish an anti-nematode fungi resource bank. The results showed that P. massoniana exhibited higher species richness than P. elliottii, but alpha / beta index analyses indicated no significant difference in endophytic fungi diversity between the two species. Endophytic fungi in both species were predominantly composed of Ascomycota and Basidiomycota, with the relative abundance of Ascomycota being higher in P. elliottii than in P. massoniana. At the genus level, significant divergences were observed: Neoheleiosa and Catanulostroma were significantly reduced in P. massoniana, whereas Paraconiothyrium was significantly more abundant in P. massoniana than in P. elliottii. Although endophytic fungi in both species showed no significant differences in primary metabolic pathways, the enrichment level of fungal phospholipases within secondary metabolic clusters was markedly high-er in P. massoniana than in P. elliottii (P<0. 001).
