影响, 结果表明: (1) 任豆种子用 75%酒精、 0. 1%升汞分别浸泡灭菌 10, 30 min, 再用始温为 85 ℃无
菌水浸种 24 h 效果最好, 种子萌芽快, 种子发芽率可达 92. 16%; (2) RD 基本培养基中 NO
-
3 、 K
+
、 Ca
2+
含量分别以 600, 360, 330mg / L 为最优配比, 最佳继代培养基配比为 RD+6-BA 0. 2 mg / L+NAA 0. 01
mg / L, 20 d 增殖系数可达 3. 32; (3) NAA 对任豆组培芽生根以及生长有极显著的影响, 最佳生根培养
基配比为 1 / 2 MS+NAA 0. 5 mg / L+AC 1. 0 g / L, 生根率可达 100%, 且根系粗壮发达, 组培苗生长健壮,
叶片浓绿舒展。 |

In order to improve the tissue culture regeneration technology of Zenia insignis, the effects of
different contents of NO
-
3 , K
+
, and Ca
2+
in basic medium and different types and concentrations of plant growth
regulators on the proliferation and rooting of Z. insignis were investigated by means of micro-cuttings. The results showed as follows: (1) The seeds of Z. insignis were soaked with 75% alcohol for 10 min and 0. 1%
HgCl
2
for 30 min, respectively, and then soaked with sterile water at 85 ℃ for 24 h, which had the best effect,
rapid seed germination and seed germination rate can reach 92. 16%. (2) The optimal ratio of NO
-
3 , K
+
and
Ca
2+
in RD basic medium was 600 mg / L, 360 mg / L and 330mg / L, respectively. The optimal subculture medium was RD + 6-BA 0. 2 mg / L + NAA 0. 01 mg / L, and the proliferation coefficient reached 3. 32 at 20 days.
(3) NAA had a very significant effect on the rooting and growth of Z. insignis Chun. The optimal rooting medium was 1 / 2MS + 0. 5 mg / L NAA+ AC 1. 0 g / L, the rooting rate could reach 100%, and the roots were robust
and developed, the tissue culture seedlings grew robustly, and the leaves were dark green and stretched. |
