大叶相思瓶内扦插生根研究

摘要:
大叶相思 Acacia auriculiformis 于上世纪六七十年代从澳大利亚引种至我国, 经选育培养, 目 前在两广和福建地区广泛栽植, 是热带造林的重要树种, 具有广泛的应用前景。 为了快速繁殖优良大叶相 思品种, 促进大叶相思的推广和应用, 选取大叶相思当年生腋芽饱满的枝条为插条, 开展扦插抑菌剂类型 及浓度、 插条类型和消毒方式、 生根促进剂浓度及处理时间等系列研究, 以期建立一种瓶内扦插生根技 术。 研究结果表明: (1) 两种抑菌剂的效果显著不同, 百菌清抑菌效果优于多菌灵。 当培养基中含 0. 2 g· L -1 百菌清时, 插条存活率为 83. 33%, 污染率 5. 56%, 褐化率 11. 11%。 当培养基中含 0. 4 g·L -1 多菌灵 时, 插条存活率 66. 67%, 污染率 26. 37%, 褐化率 6. 94%; (2) 用 0. 8 g·L -1 多菌灵对中部带叶茎段进 行消毒处理 3 min, 茎段褐化率和污染率为 0. 00%, 存活率为 100%, 显著优于其他插条类型与消毒处理 组合; (3) 最佳生根处理为 0. 8 g·L -1 IBA 浸泡枝条 3 h, 枝条存活生根率为 100%。 综上所述, 成功建 立了大叶相思瓶内扦插生根技术体系, 实现了扦插及生根的室内培养, 极大地简化了扦插基质及管理环 节, 对大叶相思今后的推广应用具有重要意义。

In Vitro Cutting Rooting Technique of Acacia auriculiformis

Abstract:
Acacia auriculiformis was introduced to China from Australia in the 1960s and 1970s, and after selection and cultivation, it is now widely planted in Guangdong, Guangxi, and Fujian, and is an important tree species for tropical afforestation, with a wide range of application prospects. In order to rapidly propagate excellent varieties of A. auriculiformis and promote the dissemination and application of A. auriculiformis plants, branches with full axillary buds in the current year of A. auriculiformis were selected as the cuttings, and a series of researches on the types and concentrations of concentrations, the types and disinfection methods of cuttings, the concentrations and treatment time of rooting promoters were carried out to establish a rooting technology of cutting in bottles. The results were as follows; (1) The effectiveness of the two disinfectants varies significantly, with chlorothalonil demonstrating superior antibacterial properties compared to carbendazim. With 0. 2 g·L -1 chlorothalonil in the medium, the survival rate of cuttings was 83. 33%, the contamination rate was 5. 56%, and the browning rate was 11. 11%. The survival rate, contamination rate and browning rate of cuttings were 66. 67%, 26. 37% and 6. 94% when the medium contained 0. 4 g·L -1 carbendazim. (2) Treating the middle stem cuttings with leaves using 0. 8 g·L -1 carbendazim for a disinfection duration of 3 minutes resulted in a browning rate and contamination rate of 0. 00%, with a survival rate of 100%. This was significantly better than other types of cuttings and disinfection treatment combinations. (3) The optimal rooting treatment involves immersing the cuttings in a solution of 0. 8 g·L -1 IBA for 3 hours, resulting in a 100% survival and rooting rate for the cuttings. This new cutting and rooting technique system simplified the cutting substrate and can be carried out indoors, which greatly streamlines the cutting management process and holds significant potential for future promotion and application.
