嘉氏羊蹄甲组织培养体系的建立

摘要:
以嘉氏羊蹄甲 Bauhinia galpinii 带腋芽茎段为外植体,建立组织培养快繁体系。结果表明: 最适合的嘉氏羊蹄甲带腋芽茎段诱导的培养基为:MS+0.5 mg · L-1 6-BA+0.05 mg · L-1 NAA,诱导率为 80.00%;最佳增殖培养基为 :WPM+0.50 mg · L-1 6BA+0.40 mg · L-1IAA,增殖系数达 4.62;最佳生根培养 基为:1/2MS+1.00 mg · L-1 IBA +0.50 mg · L-1NAA+0.10 mg · L-1 IAA +15 g · L-1 蔗糖 +7 g · L-1 卡拉胶,生 根率达 98.00%;最佳移栽基质为:V(黄心土):V(泥炭):V(珍珠岩)= 1:3:1,试管苗移栽成活率达 86.7%。试验建立了嘉氏羊蹄甲组织培养体系,提高了嘉氏羊蹄甲增殖率、生根率和移栽成活率。

Abstract:
The tissue culture system of Bauhinia galpinii with axillary buds was established. The results showed that the impact of.the most suitable medium for the induction of stem segment with axillary bud was MS + 0.5 mg · L-1 6-BA + 0.05 mg · L-1NAA, the induction rate was 80.00%; the best proliferation medium was WPM + 0.50 mg · L-1 6BA + 0.40 mg · L-1 IAA, the proliferation coefficient was 4.62; the best rooting medium was 1 / 2 ms + 1.00 mg · L-1 IBA + 0.50 mg · L-1 NAA + 0.10 mg · L-1 IAA + 15 g · L-1 sucrose + 7 g · L-1 carrageenan, the rooting rate was 98.00%; the best transplanting medium was V (yellow soil): V (peat): V (perlite) = 1:3:1, the survival rate of plantlets transplanting was 86.7%. The tissue culture system was established, which could improve the proliferation rate, rooting rate and transplanting survival rate.
