枫香组织培养快繁育苗技术研究

摘要:
以枫香2 a生苗的嫩芽为外植体进行组织培养,分别从不同优良家系建立4个无性系,同时进行组培快繁育苗技术研究。试验结果表明,适宜外植体诱导的培养基为MS KT 0.2 mg/L NAA 0.5 mg/L CM 50 mL/L,适宜不定芽增殖的培养基为改良MS BA 1.5 mg/L NAA 0.2 mg/L。外植体萌动后适时转入增殖培养是技术关键点。不同无性系增殖培养的增殖倍数有差异,平均为3.18。最佳生根培养基为1/2改良MS+IBA 0.2 mg/L+NAA 0.2 mg/L+1.6%胺鲜酯 0.02 mg/L,试验生根率最高为87.2%,不同无性系的植株生根能力差异不大。再生植株上袋后30 d 的平均存活率接近90%。

Abstract:
This paper deals with the preliminary results on clonal tissue culture and rapid propagation of Liquidambar formosana. Isolated explants derived from elite trees of 4 families were induced to form adventitious buds on a cytokinin and auxin supplemented medium. The results from subcultures indicated that the medium of modified MS supplemented with BA 1.5 mg/L and NAA 0.2 mg/L was the optimum propagation medium for multiplication of adventitious buds, in which the multiple coefficient reached 3.18 averagely. Meanwhile, the trial also identified that the optimum medium for root induction was 1/2 modified MS supplemented with IBA 0.2 mg/L, NAA 0.2 mg/L and diethyl aminoethyl hexanoate AC (1.6%) 0.02 mg/L, and 87.2% of rooting percentage could be obtained. The plantlet survival rate was nearly 90% provided that the nursery condition was acceptable.
