黄金香柳的组织培养与快速繁殖

摘要:
本文报道黄金香柳的组织培养和快速繁殖试验。结果表明:以黄金香柳嫩茎作外植体进行培养,不定芽诱导率高,经连续3次培养不定芽增殖倍数可达7.9-10.7。适宜黄金香柳丛芽快速增殖的培养基配方为MS+6-BA0.5mg/L+NAA0.01mg/L+蔗糖30g/L,一次继代培养时,平均每瓶无菌材料可提供45.5个枝芽(≥1cm)用于转接生根。诱导植株生根的最佳培养基配方为MS+IBA1.0~1.5mg/L+蔗糖15g/L,适宜的培养时间为21~25d,生根率达96%以上。移植的试管植株90%以上存活。

Abstract:
Abstract This paper deals with the preliminary results on tissue culture and rapid propagation of Melaleuca bracteata F. Fuell. . After 3 subcultures, a large number of adventitious buds could be induced from shoot explants on MS and DCR media supplemented with 6-BA0.5~2.0mg/L and NAA0.01mg/L, on which increased the multiple buds nearly tenfold. Nevertheless, the optimum propagation medium for subculture selected from the experiments was MS containing 6-BA0.5mg/L and NAA0.01mg/L, under this procedure be able to obtain 45.5 shoots for rooting culture from one culture bottle in a period of subculture averagely. The trial also identified that the optimum medium for root induction was MS supplemented with IBA 1.0~1.5mg/L and 96% of rooting percentage was obtained. The plantlet survival rate could exceed 90% provided that the nursery condition was acceptable.
