黑木相思根蘖组培繁育技术研究

摘要:
本文以根蘖促萌和组织培养相结合的方法研究黑木相思优良无性系快繁技术,4个参试的无性系都获得了根蘖萌条,最高可达12条/根。采集10-20cm的萌条作外植体进行组培快繁时,灭菌成功率较高,最高可达80.9%。试验结果表明以根蘖萌条为外植体诱导的组培苗生根性能好,在黑木相思组培增殖中有效防止苗木发黄和促进不定芽叶片舒展是两个关键因子,通过降低培养基的含糖量是一种有效的途径,由此经3次继代培养无菌苗数量平均达23株/瓶。

The study on propagation of root sucker and tissue culture for Acacia melanoxylon

Abstract:
Abstract: The superior clones propagation of blackwood were studied by means of combinating root sucker and its tissue culture. The shoots were obtained from all the roots of four fine clones. For the best results, 12 shoots per root were introduced. Using the explants from 10-20cm length root shoots, the best sterilization rate is 80.9% in blackwood tissue culture. And new shoots from these explants have very good rooting ability. Overcoming shoots etiolation and promoting leaves development are two key factors for the subculture. Reducing medium suger concentration is necessary. The average of shoots per bottle is 23 after 3 times' subculture by this way.blackwood
