相思基因组DNA提取及其RAPD初步分析

摘要 以马占相思、大叶相思、厚荚相思幼苗的叶片为材料,采用SDS法、改良SDS法和改良CTAB法进行了基因组DNA的提取并对DNA提取方法的作了比较,结果表明改良CTAB法提取效果最好。同时利用改良CTAB法提取出的DNA为模板进行了RAPD分析,鉴别了不同品种之间的多态性。11个引物共扩增出117条谱带,树种间遗传多态性明显高于种内。

Abstract:
Abstract: The methods of SDS, modified SDS and modified CTAB were adopted to extract genomic DNA from leaf tissues of A. mangium, A. auriculiformis and A. crasscarpa. The results showed that modified CTAB method was the best method. In addition, the paper analyzed the genetic diversity of Acacia using RAPD, based on DNA which was extracted by modified CTAB method was employed as template. A total of 117 sites were detected by 11 perimers, which the proportion of polymorphic loci was 54.70%.
